ISOLASI DAN IDENTIFIKASI Streptomyces spp. PENGHASIL ENZIM KITINASE DARI LUMPUR SELOKAN
Abstrak
Streptomyces is a bacterium of the Actinomycetes group. This genus is known to produce various active compounds including antibiotics, antivirals, and enzymes. Enzymes that can be found in bacteria and fungi is one of them chitinase enzyme. The production of chitinase enzyme from bacteria is better than chitinase from other sources because of its proliferation occurs in a relatively short time. Chitinase activity of chitinolitic bacteria is potentially to be used as a biological control agent against pathogenic fungi and insect pests. This study aims to isolate, characterized and identify the chitin enzyme activity of Streptomyces spp. collected from sewage slurry. The activity of chitinase enzymes is known through the clear zone formed on the medium. Five Streptomyces species were isolated from the sewage slurry collected 3 different places. Out of 5 species, two species, Streptomyces sp.3 and Streptomyces sp.5, were able to produce chitinase enzyme, while three others were not. The species of Streptomyces sp.3 producing chitinase enzyme was performed a clear zone in chitin media with average of 1.85±0.07 cm and Streptomyces sp.5 was 0.65±0.14 cm. Therefore, Streptomyces sp.3 and Streptomyces sp.5 are a potential source of bacteria producing chitinase enzyme for a biological control agent.
